LABORATORY PREPARATION OF ZIEHL NEELSEN STAINING FOR MICROBACTRIA TUBERCULOSIS



ZIEHL-NEELSEN: This stain is used for preparation of specimen for examination of mycobacterium tuberculosis; this method used three solutions, which are;
Carbol Fuchsin
Acid alcohol
Counter stain, which could be safranin i.e. depend on the laboratory scientist choice.
Ziehl-Neelsen acid-fast staining procedure
ZIEHL NEELSEN PROCEDURE

1 Carbol Fuchsin Solution

Basic fuchsin                        10g
Phenol crystal                       45g
Ethyl alcohol                        100ml
Distilled water                      900ml

PREPARATION
Dissolved the phenol in the distilled water, leave it and dissolved basic fuchsin in the ethyl alcohol, this can be achieved by gentle heat. After that combine the two solutions and mixed leave it at room temperature overnight.
After that filter and label.
Or weight the basic fuchsin and phenol crystal into 2 liter flask and dissolved by heating on a boiling water bath. This process content should be shaken frequently until the solution is dissolved. Add alcohol mixed and add distilled water, allowed to remain at room temperature overnight filter with wet filter paper label.

2 Acid alcohols
Hydrochloric Acid                             30ml
Ethyl alcohol                                       97ml

PREPARATION
Mixed hydrochloric acid with ethyl alcohol record label and store

3 Counter Stain
Methyline blue or malachite green can be used
