PROCEDURE PREPARE THICK FILM FROM CULTURE COLONIES



  1. Sterilized wire loop by passing through flame, allowed it to red hot bring it out and allowed it to cool before picking a colony
  2. Take a loop full of culture; transfer to a clean grease free glass slide spread the sample with the loop to form a thick film of a fluid.
  3. sterilized the wire loop
  4. Leave the film to air dry without heating and pass through the flame 3 time to kill the bacteria and fixed it to a slide.
  5. the slide should be allowed to cool
    6. CULTURE COLONIE
  6. stain with the right stain
examine with microscopy, by microscopic procedure
